Assessment of the cytotoxic activity of a compound on a model of the human alveolar tissue

Human and animal health In vitro/ex vivo cytotoxicity

Background of in vitro - 3D tissue model development

  • Cell culture is an important and necessary process in drug discovery and cancer research
  • Two-dimensional (2D) cell culture has been the method used to culture cells since the early 1900s but has many limitations due to 2D models inaccurately representing tissue cells in vitro
  • 3D tissue model of the human airways EpiAlveolar (MatTek) was engineered to provide human-relevant results in an easy-to-use in vitro system
  • EpiAlveolar tissues are produced on cell culture inserts. The epithelium exhibits functional tight junctions and a robust barrier
Assessment of the cytotoxic activity of a compound on a model of the human alveolar tissue

Objectives of the model

  • To determine the putative cytotoxic activity of a molecule using a 3D tissue model of the human airways

Our approach at Vibiosphen

  • A range of concentrations of the molecule was applied on the tissue in triplicate
  • The plates were incubated at 37°C 5% CO2
  • Viable cells were quantified with Cell Titer Glo 2.0 cell viability assay

Outcomes of in vitro - 3D tissue model

Assessment of the cytotoxic activity of a compound on a model of the human alveolar tissue

  • 3D tissue models originated from human cells provide human-relevant results and therefore contribute to avoiding the late-stage failures of in vivo testing
  • 3D tissue models can be used in drug discovery, cancer research….
