Background of bacterial genome editing

• Colibactin is a genotoxin synthesized by Escherichia coli that induces DNA damage that can lead to gene mutations, chromosomal instability, and senescence.
• Colibactin-producing E. coli strains are associated with cancerous and precancerous colorectal tissues and are suspected of promoting colorectal carcinogenesis
• Polyamines are highly abundant in cancer tissue

Objectives of the model

• To inactivate genes required for the synthesis of spermidine.
• To determine the roles of polyamine spermidine in colibactin genotoxic activity

• Inactivate the genes speB, speC, speE, and speG, required for the synthesis of spermidine by using the lambda Red recombinase method

• Provide in vitro data on the production of colibactin by each above mutant through bacterium-host cell interactions
